Ca2+/Calmodulin-dependent kinase II delta B is essential for cardiomyocyte hypertrophy and complement gene expression after LPS and HSP60 stimulation in vitro BJMBR
Cruz Junho,C.V.; Trentin-Sonoda,M.; Alvim,J.M.; Gaisler-Silva,F.; Carneiro-Ramos,M.S..
Inflammation plays an important role in the development of cardiovascular diseases (CVDs), suggesting that the immune system is a target of therapeutic interventions used for treating CVDs. This study evaluated mechanisms underlying inflammatory response and cardiomyocyte hypertrophy associated with bacterial lipopolysaccharide (LPS)- or heat shock protein 60 (HSP60)-induced Toll-like receptor (TLR) stimulation and the effect of a small interfering RNA (siRNA) against Ca2+/calmodulin-dependent kinase II delta B (CaMKIIδB) on these outcomes. Our results showed that treatment with HSP60 or LPS (TLR agonists) induced cardiomyocyte hypertrophy and complement system C3 and factor B gene expression. In vitro silencing of CaMKIIδB prevented complement gene...

Comparative morpho-anatomical studies of the lesions caused by citrus leprosis virus on sweet orange Anais da ABC (AABC)
Marques,João P.R.; Kitajima,Elliot W.; Freitas-Astúa,Juliana; Appezzato-da-Glória,Beatriz.
The leprosis disease shows a viral etiology and the citrus leprosis virus is considered its etiologic agent. The disease may show two types of cytopatologic symptom caused by two virus: nuclear (CiLV-N) and cytoplasmic (CiLV-C) types. The aim of this study was to compare the morpho-anatomical differences in the lesions caused by leprosis virus-cytoplasmic and nuclear types in Citrus sinensis (L.) Osbeck 'Pêra'. Leaf and fruit lesions were collected in Piracicaba/São Paulo (cytoplasmic type) and Monte Alegre do Sul/São Paulo and Amparo/São Paulo (nuclear type). The lesions were photographed and then fixed in Karnovsky solution, dehydrated in a graded ethylic series, embedded in hydroxy-ethyl methacrylate resin (Leica Historesin), sectioned (5 μm thick),...

Cyclosporin-A does not affect skeletal muscle mass during disuse and recovery BJMBR
Aoki,M.S.; Miyabara,E.H.; Soares,A.G.; Salvini,T.F.; Moriscot,A.S..
Cyclosporin-A (CsA) is an immunosuppressive drug that acts as an inhibitor of calcineurin, a calcium phosphatase that has been suggested to play a role in skeletal muscle hypertrophy. The aim of the present study was to determine the effect of CsA administration (25 mg kg-1 day-1) on skeletal muscle mass and phenotype during disuse and recovery. Male Wistar rats received vehicle (N = 8) or CsA (N = 8) during hind limb immobilization (N = 8) and recovery (N = 8). Muscle weight (dry/wet) and cross-sectional area were evaluated to verify the effect of CsA treatment on muscle mass. Muscle phenotype was assessed by histochemistry of myosin ATPase. CsA administration during immobilization and recovery did not change muscle/body weight ratio in the soleus (SOL)...

Effect of hepatocyte growth factor and angiotensin II on rat cardiomyocyte hypertrophy BJMBR
Chen,Ai-Lan; Ou,Cai-Wen; He,Zhao-Chu; Liu,Qi-Cai; Dong,Qi; Chen,Min-Sheng.
Angiotensin II (Ang II) plays an important role in cardiomyocyte hypertrophy. The combined effect of hepatocyte growth factor (HGF) and Ang II on cardiomyocytes is unknown. The present study was designed to determine the effect of HGF on cardiomyocyte hypertrophy and to explore the combined effect of HGF and Ang II on cardiomyocyte hypertrophy. Primary cardiomyocytes were isolated from neonatal rat hearts and cultured in vitro. Cells were treated with Ang II (1 µM) alone, HGF (10 ng/mL) alone, and Ang II (1 µM) plus HGF (10 ng/mL) for 24, 48, and 72 h. The amount of [³H]-leucine incorporation was then measured to evaluate protein synthesis. The mRNA levels of β-myosin heavy chain and atrial natriuretic factor were determined by real-time PCR to evaluate...

Castoldi,Robson Chacon; Aleixo,Paulo Henrique; Pereira,Alysson Christian Jesus; Ferreira,Sergio Ruiz; Garcia,Thiago Alves; Ozaki,Guilherme Akio Tamura; Koike,Tatiana Emy; Júnior,Danilo Antônio Corrêa Pinto; Seraphim,Patrícia Monteiro; Belangero,William Dias; Filho,José Carlos Silva Camargo.
To analyze the effects of concurrent training (CT) on the muscle fibers of Wistar rats submitted to standard and hypercaloric diets. In total, 40 rats were used, divided into 4 groups: Sedentary Group (GS); Exercise Group (GE), Obese Sedentary Group (OS) and Obese Exercise Group (OE). The animals performed a CT protocol consisting of: muscle strength training and aerobic training, carried out 3 times a week for 45 days. The smallest diameter of muscle fibers (MDF) was analyzed to evaluate muscle hypertrophy. It was observed that the OE group presented a significant decrease in MDF, compared to the OS group (OE=77.41 µm vs. OS=98.58 µm). In addition, the animals that performed CT demonstrated muscle hypertrophy (GE=74.39 µm vs. GS=72.13 µm). In conclusion,...

Fasting: a major limitation for resistance exercise training effects in rodents BJMBR
das Neves,W.; de Oliveira,L.F.; da Silva,R.P.; Alves,C.R.R.; Lancha Jr.,A.H..
Protocols that mimic resistance exercise training (RET) in rodents present several limitations, one of them being the electrical stimulus, which is beyond the physiological context observed in humans. Recently, our group developed a conditioning system device that does not use electric shock to stimulate rats, but includes fasting periods before each RET session. The current study was designed to test whether cumulative fasting periods have some influence on skeletal muscle mass and function. Three sets of male Wistar rats were used in the current study. The first set of rats was submitted to a RET protocol without food restriction. However, rats were not able to perform exercise properly. The second and third sets were then randomly assigned into three...

Fiber Type Composition in Semitendinous Muscle of Wistar Rats and Effects of Intermittent Training on its Hypertrophy International Journal of Morphology
Ferreira,Alexandre; Campos,Gerson Eduardo Rocha.
Skeletal muscles respond to several stimuli changing their phenotype. Muscular fibers adaptation capability is related to the presence of several myosin heavy chains (MHC). These express four types of pure fibers: I, IIA, IID and IIB containing MHCI, IIa, IId and IIb, respectively. Among pure fibers, there are hybrid fibers, which can express two or more types of myosins. In this study, types of fibers constituting male Wistar rats semitendinous and their myosin heavy chains, as well as influence of intermittent training on hypertrophy of these fibers have been checked through MATPase histochemical technique and electrophoretic proteins separation. All types of pure and hybrid muscular fiber have been found, however the fibers of the types IIA, IID and IIB...

Impaired beta-adrenergic response and decreased L-type calcium current of hypertrophied left ventricular myocytes in postinfarction heart failure BJMBR
Saraiva,R.M.; Chedid,N.G.B.; Quintero H.,C.C.; Díaz G.,L.E.; Masuda,M.O..
Infarct-induced heart failure is usually associated with cardiac hypertrophy and decreased ß-adrenergic responsiveness. However, conflicting results have been reported concerning the density of L-type calcium current (I Ca(L)), and the mechanisms underlying the decreased ß-adrenergic inotropic response. We determined I Ca(L) density, cytoplasmic calcium ([Ca2+]i) transients, and the effects of ß-adrenergic stimulation (isoproterenol) in a model of postinfarction heart failure in rats. Left ventricular myocytes were obtained by enzymatic digestion 8-10 weeks after infarction. Electrophysiological recordings were obtained using the patch-clamp technique. [Ca2+]i transients were investigated via fura-2 fluorescence. ß-Adrenergic receptor density was...

Morphological, anatomical and biochemical studies on the foliar galls of Alstonia scholaris (Apocynaceae) Rev. Bras. Bot.
Albert,Susy; Padhiar,Amee; Gandhi,Dhara; Nityanand,Priyanka.
Morphological, anatomical and biochemical alterations in foliar galls of Alstonia scholaris R. Br. induced by the insect Pauropsylla tuberculata (Psyllidae) are described and quantified. Galls occur isolated or agglomerated on the abaxial surface of the leaf. The insect along with the egg deposits some physiologic fluid which act as a stimulant for the induction of the gall. This stimulus brings about hypertrophy followed by hyperplasia of cells next to the location of the deposited eggs. The psyllid presents three nymphal instars, from eclosion of the egg to the adult. Hyperplasia in the palisade cells is very distinctly noticed. Hypertrophy followed by hyperplasia takes place and brings about elevation of hypodermal and palisade parenchyma which...

Morphological and morphometric analysis of skeletal muscle between male and female young adult Colossoma macropomum (Characiformes: Serrasalmidae) Neotropical Ichthyology
Mello,Fernanda De; Felippe,Daruzi; Godoy,Leandro C.; Lothhammer,Nívia; Guerreiro,Luis R. J.; Streit Jr.,Danilo P..
ABSTRACT This study aimed to evaluate muscle organization in tambaqui in order to describe the muscle growth process. We analyzed the morphometric pattern of fibers from white muscle of young-adults (300 days) by smaller diameter. The organization of white muscle exhibited a typical morphological pattern found in other fish species. Heavier animals showed higher frequency of larger diameter fibers (>50 μm ) and smaller animals had higher frequency of smaller diameter fibers (<20 μm ) (P =0.005). However, both animals showed the same frequency of intermediate diameter fibers (20-50 μm ). Body weight showed a positive correlation with muscle diameter fiber (r=0.45), being 20-50 μm the diameters that contributed the most to animal weight (P <0.0001)....

The Morphological Effects of Asthma, As Well As Conventional and Alternative Asthma Therapies on Parietal and Chief Cells in the Stomach of BALB/c Mice International Journal of Morphology
Vieira,Warren Antonio; Oberholzer,Hester Magdalena; Pretorius,Etheresia.
Scientific literature, although limited in this area, supports the hypothesis that asthma, by means of selective leukocyte trafficking between the various mucosal and glandular sites of the body, can have the same pathophysiological effects on the stomach as the airways. This study aimed to determine if asthma, in the absence and presence of various asthma therapies (Hydrocortisone and Modul8TM), imparted any morphological alteration on the stomach parietal and chief cells. The BALB/c murine asthmatic mouse model was the model of choice in this study. The asthma induction protocol as well as the asthma therapies were proved to be effective with the aid of bronchial lavage fluid leukocyte quantification. Fundic and pyloric biopsies were extracted at...
